The circadian clock regulates vital aspects of physiology including protein synthesis and oxidative stress response. In this investigation, we performed a proteome-wide scrutiny of rhythmic protein accrual in Drosophila melanogaster on exposure to rotenone, rotenone + hesperidin and hesperidin in D. melanogaster. Total protein from fly samples collected at 6 h intervals over the 24 h period was subjected to two-dimensional gel electrophoresis and mass spectrometry. Bioinformatics tool, Protein ANalysis THrough Evolutionary Relationships classification system was used to the determine the biological processes of the proteins of altered abundance. Conspicuous variations in the proteome (151 proteins) of the flies exposed to oxidative stress (by rotenone treatment) and after alleviating oxidative stress (by hesperidin treatment) were observed during the 24 h cycle. Significantly altered levels of abundance of a wide variety of proteins under oxidative stress (rotenone treatment) and under alleviation of oxidative stress (rotenone + hesperidin treatment) and hesperidin (alone) treatment were observed. These proteins are involved in metabolism, muscle activity, heat shock response, redox homeostasis, protein synthesis/folding/degradation, development, ion-channel/cellular transport, and gustatory and olfactory function of the flies. Atualmente são crescentes as publicações apontando a composição da dieta como um fator que pode afetar o tempo de vida e o envelhecimento dos seres vivos, no entanto ainda não são claras as relações entre proporções adequadas de macronutrientes na dieta. O objetivo principal desse estudo foi validar em nosso laboratório o método Continuos Liquid Feeding (CLF) para alimentação de Drosophila melanogaster e verificar como diferentes concentrações de macronutrientes afetam o tempo de vida do inseto. Desenvolvemos os aparatos para CLF, aplicamos dietas com diferentes concentrações de carboidrato: proteína e avaliamos seus efeitos na longevidade e resistência ao frio e fome. Tomados em conjunto nossos dados sugerem que interações metabólicas associadas ao aumento no consumo de açúcar por Drosophila melanogaster afetam sua capacidade de sobreviver em condições críticas.

The frequent use of insecticides has led to the development of insecticide resistance by many insects. In some insect populations, resistance can be developed to multiple pesticides. Up to this, insecticides resistance became a key challenge, and the amounts of the used pesticides must be as lower as possible. In this study, we measured the potency of multiple emulsions on the mortality of Drosophila virils and the ability of the fly to ingest and excrete the emulsions as a new strategy to avoid the great use of pesticides. The obtained data showed that the value of the lethal concentration of half the number of individuals decreases when the exposure duration increases, which demonstrates the sensitivity of the larvae to the toxic effect of the formulation. This can help to suggest that this type of material can be more effective and lasting effect for as long as possible. Moreover, the ingestion and excretion of the formulated product in the hind-gut can help to suggest the ability of the studied formulation to pass through the peritrophic membrane and the midgut acidic regions as two important barriers. In conclusion, the formulated microcapsule has a satisfactory insecticidal potency against D. virilis in a dose-dependent manner which might help to avoid the developed resistance to commonly used insecticides.
