Continuous in Vivo Monitoring of Amino Acid Neurotransmitters by Microdialysis Sampling with Online Derivatization and Capillary Electrophoresis Separation

Abstract: A separation-based biosensor has been developed that is capable of near-real-time analysis of aspartate and glutamate with a temporal resolution of less than 2 min in anesthetized or awake, freely moving animals. The instrument consists of a microdialysis sampling system, an on-line reactor, an injection interface, and a CE-LIF system. Primary amine analytes are derivatized with NDA/CN following microdialysis sampling using an on-line reactor to produce fluorescent CBI derivatives. The reaction takes approxima… Show more

“…It is necessary to have at least 16-18 h of incubation to obtain stable and well detected FITC derivatives [29,30]. CE-LIFD analysis of FITC derivatives gives subattomolar determination of AA mixture, which represents few thousands of injected molecules [14,27,29,39,40].…”

“…This technical improvement of the microdialysis and CE coupling requires very short derivatization periods. Therefore, it is critical to use derivatizing agents such as OPA [52][53][54] or NAD [39,55,56] to produce the fluorescent derivatives in reaction times as short as a minute. This it is not possible using FITC.…”

“…5,6 A number of methods have already been proposed for measuring L-glutamate, including chromatographic technique 7 and capillary electrophoresis. 8,9 However, these methods require time for derivatization and column separation.…”
