Continuous Flow Chemistry: A Novel Technology for the Synthesis of Marine Drugs

Laura Peña,
Paula González-Andrés,
Lucía Parte
et al.

Abstract: In this perspective, we showcase the benefits of continuous flow chemistry and photochemistry and how these valuable tools have contributed to the synthesis of organic scaffolds from the marine environment. These technologies have not only facilitated previously described synthetic pathways, but also opened new opportunities in the preparation of novel organic molecules with remarkable pharmacological properties which can be used in drug discovery programs.

“…With the application of green chemistry, biocatalysis, and computer-aided drug design, more efficiency has been gained in the synthesis of highly complex natural compound. Many difficulties, such as many chiral centers, complex synthesis steps, easily formed isomerism, and chemical agent pollution, will be solved or considerably improved by the use of the above technology (Wang et al, 2019;Chakrabarty et al, 2021;Peña et al, 2023).…”

“…On the other hand, the ability of transition metal complexes to change easily between many oxidation states makes them very convenient for the development of new catalytic pathways. These reactions usually require low catalyst loadings (sometimes less than 1% or even traces), and these are very valuable to achieve challenging couplings and provide new opportunities in terms of (i) chemo-and regioselectivity, which can be tuned by substituting with other suitable transition metal or modifying the electronic and steric properties of the ligands; (ii) asymmetric catalysis, by using chiral transition metal complexes or additional ligands as chiral auxiliaries; and (iii) reactivity, by accessing new chemical space not accessible by alternative methodologies [5].…”

“…[1] Coumarin and its derivatives possess a wide range of pharmacological activities like antitumor, anti-HIV, antibacterial, anticoagulant, anticonvulsant, antiprotozoal, antimicrobacterial, antimutagenic, anti-psoriasis, anti-inflammatory, CNS-stimulants etc. [2][3][4][5][6][7][8] Among the fused coumarin system, furocoumarin is a fusion of furan ring with coumarin in either 2,3-or 3,2-arrangement, at the c, f, g or h bonds of the coumarin system Figure 1. These fused coumarins are known to have important active pharmaceutical ingredients (API).…”
