Continuous desalting of refolded protein solution improves capturing in ion exchange chromatography: A seamless process

Abstract: Truly continuous biomanufacturing processes enable an uninterrupted feed stream throughout the whole production without the need for holding tanks. We have utilized microporous anion and cation exchangers into which only salts, but not proteins, can penetrate into the pores for desalting of protein solutions, while diafiltration or dilution is usually employed for feed adjustments. Anion exchange and cation exchange chromatography columns were connected in series to remove both anions and cations. To increase … Show more

“…It was shown that IB processing can be boosted when operating the notorious refolding step in a continuous mode (Wellhoefer et al, 2014; Walch and Jungbauer, 2017). Within different studies, the continuously refolded lysozyme exhibited higher refolding yields than commonly performed batch/fed-batch refolding (Farshbaf et al, 2002).…”

“…Klutz et al reported the integration of different downstream processing steps using single‐use equipment, while Shamashkin et al reported the integration of a protein A capture step and a flow‐through anion‐exchange (AEX) step. Regarding integrated downstream operations, Hughson et al reported a straight‐through purification strategy combining chromatography and membrane steps while Walch and Jungbauer reported continuous desalting with subsequent ion exchange capture. Conversely, Andersson et al discussed design and control strategies for a variety of different integrated chromatographic operations.…”
