Continuous and Batch Production of Chloroperoxidase by Mycelial Pellets of Caldariomyces fumago in an Airlift Fermentor

Abstract: Batch and continuous production of the extracellular heme glycoprotein chloroperoxidase (CPO) was studied with an airlift fermentor. We induced Caldariomycesfumago CMI 89362 to form pellets by transferring a small inoculum volume in preculture prior to growth in a 1-liter fermentor. Continuous replacement of the fructose-salts medium (dilution rate, 0.008 h-l) supported continuous CPO formation at an average concentration of 128 + 10 mg of CPO liter-' for 8 days. Optimum CPO production rates averaged 1.2 + 0.1… Show more

“…The MCD was synthesized as described by Hager et al [12] and molar extinction coefficient was determined to be 1.21 × 10 4 L mol −1 cm −1 . Pigment absorption was measured spectroscopically at 575 nm in 20 mM potassium phosphate buffer pH 5.5 [16]. Determination of protein concentration was performed spectrophotometrically at 595 nm as described by Bradford [17] using bovine serum albumin as standard.…”

“…Like most peroxidases, CPO catalyzes the peroxide-dependent oxidation of a wide spectrum of organic and inorganic compounds (24). CPO can be produced by C. fumago grown in a defined fructose-salts medium, in a semicontinuous or continuous manner, and at up to 0.5 to 0.8 g of enzyme per liter (2,5,6). Under these conditions, it is the only protein secreted and is readily purified from the culture medium.…”
