Context-specific regulation of surface and soluble IL7R expression by an autoimmune risk allele

Abstract: IL-7 is a key factor in T cell immunity and common variants at IL7R, encoding its receptor, are associated with autoimmune disease susceptibility. IL7R mRNA is induced in stimulated monocytes, yet a function for IL7R in monocyte biology remains unexplored. Here we characterize genetic regulation of IL7R at the protein level in healthy individuals, and find that monocyte surface and soluble IL7R (sIL7R) are markedly induced by lipopolysaccharide. In monocytes, both surface IL7R and sIL7R expression strongly ass… Show more

“…Though IL7R had originally been reported to be essential exclusively for lymphocyte development and survival, a few recent studies, including ours, have found that it also plays roles in myeloid cell development [6][7][8] . Interestingly, in addition to our finding that IL7R regulates trMac specification during mouse fetal development 4 , studies in humans have reported that other myeloid cell types, including monocytes and eosinophils, may express IL7R 7,9 . Furthermore, these cells can upregulate IL7R mRNA and surface protein upon stimulation with lipopolysaccharide (LPS), a known activator of eosinophils.…”

“…Our data presented here reveal that IL7R is required for specific myeloid cell homeostasis in the lungs, PB, and BM of adult mice. Other groups have reported that human eosinophils have detectable levels of IL7Ra mRNA and surface protein 7,8 , and that IL7R mRNA can be induced in human monocytes with LPS stimulation 9 . Similarly, we previously found that trMacs in the lung and other tissues transiently express IL7Rα during development 6 .…”

“…On the other hand, a highlighted biological process detected in the CCB breed was the negative regulation of T-cellmediated cytotoxicity, associated with IL7R gene. In humans, variations in this gene have been linked with autoimmune risk (Al-Mossawi et al, 2019), while in livestock animals, it has been described to be associated with growth traits (Jia et al, 2019;Zhu et al, 2019), which corroborates the purpose of this breed formation as a beef cattle.…”

“…Furthermore, we performed the overall survival analysis of 663 immune-related genes and identified that 600 genes (90.5%) were associated with better outcomes in metastatic melanoma patients. PPI network and two significant models were constructed among them and highly connected nodes in the models, including IL7R, CD38 and CXCR4 were closely associated with immune response, especially participating in regulating CD4+ and CD8+ T cells [22][23][24].…”
