Construction of SSR-based chromosome map in bunching onion (Allium fistulosum)

Abstract: We have constructed a linkage map of bunching onion (Allium fistulosum L., 2n = 16) using an F(2) population of 225 plants. The map consists of 17 linkage groups with 212 bunching onion SSR markers and 42 bulb onion (A. cepa L.) SSR, InDel, CAPS or dCAPS markers, covering 2,069 cM. This is the first report of a linkage map mainly based on SSR markers in the genus Allium. With the 103 anchor markers [81 bunching onion SSRs, 11 bulb onion SSRs and 11 bulb onion non-SSRs (1 InDel, 9 CAPSs and 1 dCAPS)] whose chro… Show more

“…However, the degree of genetic trait expression depends on the interaction of genotype with environment and farming practices. With this respect, Tsukazaki et al (2008) constructed a new linkage map on Allium fistulosum, the first one mainly based on SSR markers in the genus Allium, which will be useful for genetic studies such as QTL analysis of agronomic traits.…”

“…SSR markers were selected to be evenly distributed across each linkage group of the bunching onion map (1-4 loci per linkage group) (Tsukazaki et al 2008). Of these, ACM096, ACE010 and ACE044 are bulb onion ESTderived SSR markers (Kuhl et al 2004, Tsukazaki et al 2008, and AMS14 is a bulb onion genomic SSR marker developed by Fischer and Bachmann (2000).…”

“…This QTL is thought to be constantly effective on seedling growth in the present population, showing clear overdominance, as discussed below. Recently, Tsukazaki et al (2008) constructed an SSR-based bunching onion map and assigned the linkage groups to chromosomes of bunching onion and/or bulb onion (Allium cepa L.) using alien monosomic addition lines developed by Shigyo et al (1996). It is suggested that the linkage group D3 corresponds to linkage group 2a on the SSR-based map, which is assigned to A. cepa chromosome 2 (Tsukazaki et al, 2008).…”
