Construction of mini-Tn10luxABcam/Ptac-ATS and its use for developing a bacteriophage that transduces bioluminescence to Escherichia coli O157:H7

Abstract: Mini-Tn10luxABcam/Ptac-ATS was constructed in order to develop a luciferase-transducing bacteriophage for detecting Escherichia coli O157:H7. The transposon was designed to deliver a 3.6-kb insertion that confers n-decanal-dependent bioluminescence and resistance to chloramphenicol and was constructed using mini-Tn10cam/Ptac-ATS in the plasmid pNK2884 and luxAB from Vibrio harveyi. xV10, a temperate bacteriophage infecting common phage types of Escherichia coli O157:H7, was mutagenized as a prophage in E. coli… Show more

“…Although phage V10 has been shown to be specific to E. coli O157, it does not plaque on all O157 strains (30). On the other hand, AVR2-V10 killed all 49 O157 isolates tested and may therefore provide complete coverage within the serotype.…”

“…Bacteriophage V10 is a phage of the family Podoviridae that specifically infects E. coli O157 strains (30). V10 is similar to P22-like phages, particularly phage ε15 (8,12).…”

“…In contrast, use of stable chromosomally lux-marked constructs, such as that reported in this study, eliminates the requirement of applying a selective pressure and is thus desirable for long-term survival experiments. Although the construction of a chromosomally lux-marked E. coli O157:H7 strain has been reported previously (41), this was a luxAB mutant, which is dependent on addition of n-decanol for display of the luminescent phenotype. The construct described in this study alleviates this need for additions prior to analysis.…”

“…Recently, a renewed interest in phage therapy has surfaced with the advent of antibiotic resistance (3). Phages have also been found useful as diagnostic tools for rapid detection of such pathogens as Mycobacterium tuberculosis, Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp., and Staphylococcus aureus (5,28,29,36). These diagnostic methods make use of phages that have been genetically modified to carry a reporter gene.…”
