Construction of an immune‐related LncRNA signature with prognostic significance for bladder cancer

Abstract: Bladder cancer (BLCA) is the most common urinary malignancy in the United States, and approximately 814,00 new cases and 17,980 deaths have been recorded in the 2020s. 1 There are two major subtypes: non-muscle-invasive bladder cancer (NMIBC) and muscleinvasive bladder cancer (MIBC). Currently, clinicians prefer to undertake transurethral resection of the bladder tumour followed by intravesical instillations of chemotherapy or immunotherapy as the therapeutic approach for NMIBC. 2,3 Radical cystectomy remains

“…Analysis of the public dataset (TCGA-BLCA cohort and GSE89006 in GEO database) suggested that RP11-89 was upregulated in BLCA and closely associated with prognosis of BLCA patients in the previous study [ 25 ]. As shown in Fig.…”

“…The immune microenvironment is critical in BLCA treatments, especially in immunotherapies such as BCG instillation and immune checkpoint inhibitor [42,43]. We previously identified a novel immune related lncRNA signature with prognostic significance for BLCA and RP11-89, one of lncRNAs in the signature, was demonstrated to play an oncogenic role through inhibiting regulated cell death [25]. Chen et al demonstrated that the underlying mechanism for ferroptosis in immune regulation could be explained by immune surveillance, which indicated that tumor cell ferroptosis and tumor suppress could be triggered or enhanced by immune interventions [13].…”

“…LncRNAs have been con rmed to be in relation to cancer immunity regulation as well as tumor microenvironment in bladder cancer 17 . Several immune-related lncRNA models have been constructed in published literature 14,18,19 . Nevertheless, these signature models are developed on the basis of expression quanti cations of immune-related lncRNAs.…”

“…Linc00936/miR-425-5p/ZC3H12A axis functioned to promote cytokine induced killer cell cytotoxicity and suppress immune escape by decreasing the contents of immunosuppressive factors VEGF, IL-10, and TGF-β1 in GC ( Li et al, 2021 ). Moreover, immune-related lncRNA (IRlncRNA) can be used to establish a model to predict prognosis in cancers ( Chen et al, 2020 ; Luo et al, 2021 ; Wang et al, 2021 ; Zhao K. et al, 2021 ). However, due to difference in data processing, the absolute expression level of genes and lncRNAs between different datasets cannot be compared directly, and appropriate normalizations and standardizations of the lncRNA expression levels are required.…”
