Construction of an Affordable and Easy-to-Build Zebrafish Facility

Abstract: In vivo biomedical research is pivotal to translate in vitro findings into clinical advances. Small academic institutions with limited resources find it virtually impossible to build and maintain typical rodent facilities for research. Zebrafish research has been demonstrated to be a valuable alternative for in vivo research in pharmacology, physiology, development and genetic studies. This article demonstrates that a functional zebrafish facility can be built in an easy and affordable manner. We demonstrate t… Show more

“…Wild-type (WT) TAB strain adult zebrafish of mixed sex were obtained from the Garvan Institute of Medical Research (Sydney, Australia) and used as parents to produce experimental fish. Adult fish were housed in clear plastic tanks (350 × 200 × 260 mm) within a custom-built zebrafish recirculating system [ 23 ]. Tanks were maintained at 28.5 °C unless otherwise specified, which was monitored using temperature data loggers (Hobo MX2201, OneTemp, Australia).…”

“…Such model organisms allow for drug safety tests at an organism level and are potentially more relevant and less resource consuming 97,98 . The maintenance of zebrafish for drug screening is relatively more cost‐effective than the use of expensive fetal bovine serum required for cell culture–based alternative models, and which is commonly extracted via cardiac puncture from bovine fetuses performed without anesthesia 99‐102 . Therefore, in the process of replacing animals for drug safety, we may inadvertently increase the demand for cardiac punctures on bovine fetuses to extract fetal bovine serum.…”

“…If water parameters, such as alkalinity, hardness, and pH are not monitored and corrected as needed, filter efficiencies will fluctuate over time. Examples of recirculating systems are the open-design solutions proposed by Burg et al [39], Paige et al [40], and Nema and Bhargava [41,42].…”
