Construction, characterization, and immunization of nanoparticles that display a diverse array of influenza HA trimers

Abstract: Current influenza vaccines do not elicit broadly protective immune responses against multiple strains. New strategies to focus the humoral immune response to conserved regions on influenza antigens are therefore required for recognition by broadly neutralizing antibodies. It has been suggested that B-cells with receptors that recognize conserved epitopes would be preferentially stimulated through avidity effects by mosaic particles presenting multiple forms of a variable antigen. We adapted SpyCatcher-based pl… Show more

“…SpyCatcher003-mi3 nanoparticles ( 80 ) were expressed in BL21 (DE3)-RIPL Escherichia coli (Agilent) transformed with the pET28a His6-SpyCatcher003-mi3 gene (Addgene) as described ( 34 , 82 ). Briefly, transformed bacterial cell pellets were lysed in the presence of 2.0 mM PMSF (Sigma).…”

“…Finally, the I53-50 NP was coassembled from 20 trimeric I53-50A and 12 pentameric I53-50B subunits to form an icosahedral nanoparticle ( 48 , 49 ). These three nanoparticles were found to be safe, stable, and easy to produce in previous studies ( 32 , 50 52 ).…”
