Induction of interferon-fl (IFN-fl) in human (BG-9), simian (CV-1) and mouse (L-929) cell lines by Sendai virus and by poly(rI), poly(rC) has been studied for its possible dependence on protein kinase C (PKC) through the use of pharmacological inhibitors (K252a and H-7) of PKC. Exposure of BG-9, CV-1 or L-929 cells to K252a (>10.025 ~tM), a staurosporine derivative, 24 h before or after induction of IFN with poly(rl).poly(rC), inhibited by > 95 % the production of IFN-fl. In contrast, virus-induced 1FN production was enhanced threefold or more by K252a in BG-9 and L-929 but not in CV-1 cells. A naphthalene sulphonamide inhibitor of PKC, H-7, at >/5 ~tM, decreased poly(rI).poly(rC)-induced IFN production in BG-9 and CV-1 cells by 75 to 94%, but had no effect on IFN production in L-929 cells. Viral induction of IFN was not affected significantly by H-7 in BG-9, CV-1 and L-929 cells. In contrast to these results, the calmodulin inhibitor, trifluoperazine (5 to 15 ~tM) did not affect IFN-fl production by poly(rI).poly(rC) but significantly enhanced IFN production by Sendai virus in both human and murine cell lines. Thus, in human and simian fibroblasts the induction of IFN-fl by poly(rI).poly(rC) appears to be PKC-dependent, whereas viral induction of IFN-fl is not. Results with K252a implicate PKC in non-viral induction of IFN in mouse fibroblasts, as well. Direct measurements of PKC activity in BG-9 cells exposed to several concentrations of K252a showed that the membrane PKC activity is significantly more sensitive to inhibition by K252a than is cytosolic PKC activity. In L-929 cells, K252a inhibited membrane PKC activity similarly, but was less effective as an inhibitor of cytosolic enzyme activity than in BG-9. These studies support an intregral role for PKC activity, particularly membraneassociated activity, in non-viral [poly(rI).poly(rC)] induction of IFN-fl in human, simian and mouse fibroblasts.

Y (immune) interferon (IFN-'y) was induced in cultures of fresh human lymphocytes by combined treatment with a phorbol ester (12-0-tetradecanoylphorbol 13-acetate, TPA) and the T cell mitogen phytohemagglutinin (PHA). Compared to the IFN-y yields obtained with PHA induction alone, the inclusion of TPA caused a significant enhancement of IFN-y production.Poly(A)-containing mRNA was isolated from mononuclear cells induced with TPA and PHA. Injected into Xenopus laevis oocytes, this mRNA preparation gave rise to IFN activity with characteristic properties of human IFN-y. Sucrose density gradient centrifugation analysis showed that IFN-y mRNA sedimented at 15 S, suggesting that it contains a total of about 1400 nucleotides.As a result ofrecent progress in the study ofvarious interferons (IFNs), all known IFNs can be grouped into three major classes designated IFN-a ("leukocyte"), 3 ("fibroblast"), and IFN-'y ("immune"). Identification of the three major IFN classes is based primarily on antigenic differences (1). Additional distinguishing properties include differences in various physicochemical and biological characteristics (2).Several earlier studies showed that mRNA for IFN-a and IFN-f3 can be extracted from cells actively producing IFN. Such IFN mRNA preparations can direct synthesis of biologically active IFN molecules with corresponding specificity when introduced into heterologous animal cells (3), suitable cell-free systems (4-6), or Xenopus laevis oocytes (6-8). Recent analysis of human chromosomal gene bank DNA revealed the existence ofat least eight related but distinct IFN-a sequences, differing from each other in about 10-30% of their predicted amino acid residues (12). In contrast, only one subclass ofIFN-,BmRNA has been tentatively identified in addition to the IFN-,f cDNA clone originally isolated (13).Studies of isolated IFN-a and IFN-fB DNA clones and of the corresponding IFN polypeptides produced in bacteria have already provided a wealth ofinformation about the structure and biological properties ofthese IFNs. In addition, cloning ofthese IFN genes in Escherichia coli promises to facilitate the production of large quantities of biologically active IFN.In contrast, much less information is available about IFN-y.
