Construction and evaluation of a prognosis lncRNA model for hepatocellular carcinoma

Abstract: Current studies indicate that long non‐coding RNA (lncRNA) is often abnormally expressed in hepatocellular carcinoma (HCC). We intend to generate a multi‐lncRNA signal to improve the prognosis of HCC. By analyzing 12 pairs of HCC and adjacent normal mucosal tissues, 3900 differentially expressed lncrnas were identified as candidate biomarkers for the prognosis of HCC. Then, the 12‐lncrna signature was constructed using the LASSO Cox regression method and verified in the TCGA training dataset. Finally, we estab… Show more

“…To further illustrate the performance of GILncSig in our study, we recruited two recently published lncRNA signatures for survival prediction of HCC patient. Li’s study included 12-lncRNA signature (LilncSig) and Ma’s study included 4-lncRNA signature (MalncSig) [ 28 , 29 ]. For LilncSig, Li et al analyzed 12 pairs of HCC and adjacent normal mucosal tissues and identified 3900 differentially expressed lncRNAs as candidate biomarkers for the prognosis of HCC [ 28 ].…”

“…According to previous reports, high expression levels of collagen type XXIV alpha 1 chain ( 43 ), ubiquilin 2 ( 44 ), sushi, von Willebrand factor type A, EGF and pentraxin domain containing 1 ( 45 ), acyl-CoA synthetase long chain family member 4 ( 46 ) and YTH N6-methyladenosine RNA binding protein 1 ( 47 ) are associated with the progression of HCC. Another study has demonstrated that in addition to the American Joint Committee on Cancer staging system, a long non-coding RNA-based risk score is another important factor affecting the overall survival of patients with HCC ( 48 ). However, none of the above methods are currently widely used in the clinic.…”

“…Long non-coding RNAs (lncRNAs) are special RNA molecules without protein-coding potential and are longer than 200 nucleotides [4][5][6] . It has been reported that they are important in tumorigenesis and progression [4,[7][8][9][10][11][12][13] . For example, lncRNA MiR4435-2HG overexpression has been associated with poor prognosis in colorectal cancer (CRC) [14] .…”
