An effective vaccine against Edwardsiella tarda has not been reported in substitution for high concentration of formalin for the prevention of edwardsiellosis disease. In this study, the efficacy of inactivated E. tarda was evaluated and compared by intraperitoneal (IP) injection-immunization or challenge against Japanese eel (Anguilla japonica). Formalin, formalin with heat, citric acid, pressure and electric current were used for inactivation of the bacteria, and the relative percent survival (RPS) values of pressure (600 psi for 5 min) killed cells was determined. PKC-inactivated vaccine showed-89-93 protection that was higher than others. PKC-inactivated vaccine at a concentration of 10 6 cells/fish was sufficient to induce high protection (RPS>89). Protection of the different-inactivated vaccines was evaluated at different time post immunization, and the peak of protection was observed at 9 days post-challenge. Fish immunized with PKC showed significantly (P<0.05) higher serum and mucus antibody titers elicit both systemic and mucosal adaptive immune responses, and induce specific humoral immune responses in eel. Coincident with higher protection, sera of fish immunized with the PKC vaccine had higher agglutination titers than FKC, FHKC, CAKC and ECKC. All these data strongly suggested that PKC vaccine is an effective strategy to protect eel against edwardsiellosis.

Edwardsiella tarda EIB202, a Gram-negative pathogen with strong virulence, is an opportunistic pathogen capable of causing edwardsiellosis with high mortality to fish. Alternative sigma factor 54 (RpoN) is an important regulator of virulence and stress resistance genes in many bacterial species and mainly responsible for transcription of genes in nitrogen utilization. In this study, the in-frame rpoN deletion mutant was constructed to analyze the function of RpoN in Edwardsiella tarda firstly. Compared to the wild-type and complemented strain rpoN (+), the ΔrpoN was impaired in terms of the ability to survive under oxidative stress, osmotic stress and acid resistance, as well as the growth in Luria-Bertani medium, demonstrating essential roles of RpoN in stress resistance and nitrogen utilization. In addition, the ΔrpoN displayed markedly decreased biofilm formation and chondroitinase activity and was attenuated in virulence reflected in the increased median lethal dose value and extended infection cycle. Real-time polymerase chain reaction revealed that the expression levels of σ(70) class changed in varying degrees in the rpoN mutant. Especially, the expression levels of rpoS and fliA were down-regulated 4.1-fold and 7.9-fold in stationary phase in comparison with the wild type, respectively. Furthermore, two differential expression genes, znuA and flhC, were detected in the wild type and ΔrpoN using the method of differential display reverse transcription PCR.

(2017) Transcriptomic dissection of the horizontally acquired response regulator EsrB reveals its global regulatory roles in the physiological adaptation and activation of T3SS and the cognate effector repertoire in Edwardsiella piscicida during infection toward turbot, Virulence, 8:7, 1355-1377, DOI: 10.1080/21505594.2017 ABSTRACTEdwardsiella piscicida is the leading pathogen threatening worldwide aquaculture industries. The 2-component system (TCS) EsrA-EsrB is essential for the pathogenesis of this bacterium. However, little is known about the regulon and regulatory mechanism of EsrA-EsrB or about the factors that mediate the interaction of TCS with bacterial hosts. Here, our RNA-seq analysis indicated that EsrB strongly induces type III and type VI secretion systems (T3/T6SS) expression and that it modulates the expression of both physiology-and virulence-associated genes in E. piscicida grown in DMEM. EsrB binds directly to a highly conserved 18-bp DNA motif to regulate the expression of T3SS and other genes. EsrB/DMEM-activated genes include 3 known and 6 novel T3SS-dependent effectors. All these effector genes are highly induced by EsrB during the late stage of in vivo infection in fish. Furthermore, although in vivo colonization by the bacterium relies on EsrB and T3/T6SS expression, it does not require the expression of individual effectors other than EseJ. The mutant lacking these 9 effectors showed significant defects in in vivo colonization and virulence toward turbot, and, more importantly, a high level of protection against challenges by wild-type E. piscicida, suggesting that it may represent a promising live attenuated vaccine. Taken together, our data demonstrate that EsrB plays a global regulatory role in controlling physiologic responses and the expression of T3SS and its cognate effector genes. Our findings will facilitate further work on the mechanism of molecular pathogenesis of this bacterium during infection.
