Constitutive calcium entry and cancer: updated views and insights

Abstract: Tight control of basal cytosolic Ca concentration is essential for cell survival and to fine-tune Ca-dependent cell functions. A way to control this basal cytosolic Ca concentration is to regulate membrane Ca channels including store-operated Ca channels and secondary messenger-operated channels linked to G-protein-coupled or tyrosine kinase receptor activation. Orai, with or without its reticular STIM partner and Transient Receptor Potential (TRP) proteins, were considered to be the main Ca channels involved.… Show more

“…Growing evidence shows that cancer cell hallmarks are strongly regulated by ion channels including Ca 2+ , Mg 2+ , and K + , Na + channels . Thus, we further investigated the effect of extracellular Ca 2+ and Mg 2+ on the migration of prostate cancer cells DU145.…”

“…In nonexcitable cells, Ca 2+ influx is partly mediated by Ca 2+ ‐selective channels activated by intracellular Ca 2+ store depletion called store‐operated channels (SOCs), and by store‐independent Ca 2+ channels, such as ARC channels (arachidonate‐regulated Ca 2+ channels) . SOC channels are mainly composed of Orai1 subunits, but also composed of TRPC channels in some cell types, supporting less Ca 2+ ‐selective SOCE (store‐operated Ca 2+ entry) . SOCs are activated by endoplasmic reticulum (ER) Ca 2+ depletion, and regulated by STIM1, a Ca 2+ sensor mostly localized at the ER membrane .…”

“…However, its implication in calcium signaling has not been fully established . Finally, Orai1 and some TRP are also involved in constitutive Ca 2+ entry and tumor development …”
