Conserved neural circuit structure across Drosophila larval development revealed by comparative connectomics

Abstract: During postembryonic development, the nervous system must adapt to a growing body. How changes in neuronal structure and connectivity contribute to the maintenance of appropriate circuit function remains unclear. Previously , we measured the cellular neuroanatomy underlying synaptic connectivity in Drosophila (Schneider-Mizell et al., 2016). Here, we examined how neuronal morphology and connectivity change between first instar and third instar larval stages using serial section electron microscopy. We reconstr… Show more

“…Connectivity matrices have now been reported more recently for other parts of the Drosophila brain, in both the adult (Takemura et al, 2013(Takemura et al, , 2015(Takemura et al, , 2017a and larva (e.g. Schneider-Mizell et al, 2016;Gerhard et al, 2017).…”

“…Serial-section electron microscopy (EM) allows reconstruction of neural circuits with synapse resolution [14][15][16][17][18][19][20] , but low throughput makes it difficult to compare whole brain samples and comprehensively assess plasticity. EM has been applied to assess wiring differences between species 21 , sexes 22 , genotypes 23 , and ages 4,24 . But previous studies mapped partial circuits or few samples.…”

“…We also show that dendrite elaboration occurs much later than axon extension in the U motor neurons. The observation that axon outgrowth precedes dendrite outgrowth has been widely reported (Gerhard et al, 2017;Mason, 1983;Mumm et al, 2006;Ramon y Cajal, 1909), although the mechanism that sets the time of axon or dendrite outgrowth is poorly understood.…”
