Conservation of epitopes in the oligosaccharide portion of the lipooligosaccharide of Haemophilus influenzae type b

Abstract: The antigenic characteristics of the lipooligosaccharide (LOS) of Haemophilus influenzae type b (Hib) were examined in strains obtained over an extended period of time. These Hib strains were isolated from patients with systemic Hib disease in Dallas, Tex., over a 20-year period and in New York City between 1941 and 1956. The antigenic characteristics of the LOS of these Hib strains were examined by using a set of four murine monoclonal antibodies directed against epitopes present in the oligosaccharide portio… Show more

“…Hib strain DL42 is a non-0-lactamase-producing strain isolated from the CSF of a child with meningitis . This strain has been shown to be virulent in the infant rat model for Hib invasive disease and to belong to LOS antigenic group 2, which is the predominate LOS antigenic type among invasive Hib strains (40) . rrIL1 S. rrID10 was purified from the extract of Escherichia coli (HB 101) that was transformed with a rabbit 11,10-containing plasmid.…”

“…For kdtA, orf18 (kdtB ) and nusG, no transformants were obtained. Analysis of LPS from mutant strains by immunoblotting LPS molecules from wild-type and mutant strains were analysed by colony immunoblotting using a panel of monoclonal antibodies (mAbs), each specific for particular LPS epitopes (Gulig et al, 1987;Borrelli et al, 1995;Borrelli, 1996). Table 2 shows the reactivities of mAbs 4C4, 6A2, 12D9, 5G8 and 7E7 to the wild-type strain RM7004 and mutant derivatives.…”
