Conservation and divergence in the transcriptional programs of the human and mouse immune systems

Abstract: Much of the knowledge about cell differentiation and function in the immune system has come from studies in mice, but the relevance to human immunology, diseases, and therapy has been challenged, perhaps more from anecdotal than comprehensive evidence. To this end, we compare two large compendia of transcriptional profiles of human and mouse immune cell types. Global transcription profiles are conserved between corresponding cell lineages. The expression patterns of most orthologous genes are conserved, partic… Show more

“…Additionally, the therapeutic results from an immune-competent mouse model may not translate to human patients. Despite a striking conservation of transcriptional programs during murine and human immune system differentiation, several hundred immune genes diverge and their regulatory mechanisms are less well conserved (19). The ongoing technologic advances in engrafting Il2rg mutated, immunodeficient mice with functional human immune systems will likely provide greater translational confidence (20).…”

“…Recent global comparative transcriptional studies in other hematologic lineages, i.e., immune cells, have shown both conservation and divergence (18,19). As such, it can be argued that the vast extent of divergence may be due to underlying differences in immune cell function and composition (20,21).…”

“…Such relationships are naturally represented by a tree, and computational approaches that can incorporate the tree structure while identifying regulatory modules and networks have been useful in understanding evolutionary (Xie et al 2011;Roy et al 2013;Shay et al 2013) and developmental processes . CMINT is based on a previous module inference algorithm that we developed for species lineages (Roy et al 2013) with two major extensions.…”
