Connexin 50 Functions as an Adhesive Molecule and Promotes Lens Cell Differentiation

Abstract: Connexins play essential roles in lens homeostasis and development. Here, we identified a new role for Cx50 that mediates cell-cell adhesion function. Cx50 enhanced the adhesive capability of AQP0. Interestingly, the expression of Cx50 alone promoted cell adhesion at a comparable level to AQP0; however, this cell adhesive function was not observed with other lens connexins, Cx43 and Cx46. Moreover, the adhesive property occurred in both homotypic with Cx50 expressed in both pairing cells and heterotypic with C… Show more

“…To elucidate the effect of oxidative stress on lens connexin hemichannel activity, we infected chick embryo fibroblast (CEF) cells with recombinant RCAS(A) retrovirus containing FLAG-tagged wild-type Cx50 and/ or Cx50 mutants (E48K, P88S and H156N), and treated the cells with H 2 O 2 . As we reported previously, we have not detected expression of other possible connexin subtypes or the activities of connexin channels in these cells (Banks et al, 2007;Hu et al, 2017). With retroviral infection, almost all CEF cells express exogenous connexins (Gu et al, 2003;Jiang, 2001).…”

“…Primary antibodies were detected with goat anti-rabbit IgG conjugated IRDye ® 800CW and goat anti-mouse IgG conjugated IRDye ® 680RD (1:15 000 dilution) using a Licor Odyssey Infrared Imager (Lincoln, NE, USA), as previously described. 74 The band intensity was quantified by densitometry using Image J software (NIH, Bethesda, MD, USA).…”

“…Six connexins form a hemichannel, and two hemichannels from a pair of adjacent cells constitute a gap junction. Through their vital roles in cellular communication, connexins have been shown to be required for the differentiation and maturation of different organs, such as the heart, the eye, the skeleton and nervous tissue ( Moorer and Stains, 2017 ; Su et al, 2017 ; Hu et al, 2017 ). In the mammalian inner ear, connexin 26 (Cx26, encoded by the GJB2 gene) is the main connexin, and it assembles into gap junctions in all supporting cells and fibrocytes ( Ahmad et al, 2003 ; Kikuchi et al, 1995 ; Zhao and Yu, 2006 ).…”
