Conformational Prerequisites for the in vitro Inhibition of Cholate Uptake in Hepatocytes by Cyclic Analogues of Antamanide and Somatostatin

Abstract: Constitution and conformation determine the biological activity of a peptide, as demonstrated by comparative investigations of the inhibition of cholate uptake in liver cells by somatostatin, antamanide, cyclolinopeptide A and 1 and 2. The crystal structure of 1 is the same as that of 2 over a wide range and both react strongly cytoprotectively.

“…Cyclolinopeptide 1 was shown to inhibit cholate uptake into hepatocytes, potentially protecting the liver against poisoning. 7,8 Cyclolinopeptide 1 was also demonstrated to suppress the calcium-dependent activation and proliferation of Tlymphocytes by cyclophilin-dependent calcineurin inactivation. 9,10 The immunosuppressive activity of cyclolinopeptide 1, described by others, may be partially or wholly explained by this observation.…”

“…In in vitro studies, 1 inhibits activation and proliferation of T-lymphocyte cells [7], protects liver from specific poisonous agents [8], and suppresses immunity [7,9]. Additionally, CLs 2, 5, and 7 exhibit immunosuppressive activities [1,5].…”

“…4) Reducing side effects. Long-term treatment with octreotide is associated with an increased incidence of gallstones which may be the result of an inhibition of gall bladder motility [47] or by the inhibition of the cholate uptake by somatostatin and its derivatives [48][49][50].…”
