Conflicting results obtained by RAPD-PCR and large-subunit rDNA sequences in determining and comparing yeast strains isolated from flowers: a comparison of two methods

Abstract: Sixty-six yeast strains isolated from the nectar of various plant species in Central Europe were characterized by randomly amplified polymorphic DNA PCR (RAPD-PCR) and by sequencing of the variable D1/D2 domain of large-subunit (26S) rDNA. The usefulness of both methods for the determination and comparison of unknown ascomycetous and basidiomycetous yeast strains was compared and evaluated. The reproducibility of RAPD-PCR was shown to be low and the information obtained by this method was clearly not as precis… Show more

“…1b, inset) and the feeding behavior of the moths, which do not feed on cold nights or during the colder periods of the night (e.g., Martínez del Río and Bfflrquez 1986). Fermentation by yeasts in the nectar (e.g., Herzberg et al 2002) could also add to the basal floral CO 2 release (Drawert et al 1987), and this supplementary production of CO 2 may also diminish with the drop in temperature during the night.…”

“…The species seems to have a global and ubiquitous distribution. It was isolated from volcanic soil in Iceland (Vishniac, 2002), from soil of forest in Austria (Wuczkowski and Prillinger, 2004), as well as from floral nectar in Germany (Herzberg et al, 2002). In view of the various substrates from which it has been isolated and because of its low frequency reported here an association with grapes seems unlikely.…”
