Conditional Expression of a Dominant-Negative c-Myb in Vascular Smooth Muscle Cells Inhibits Arterial Remodeling After Injury

Abstract: Abstract-Inhibiting activity of the c-Myb transcription factor attenuates G 1 to S phase cell cycle transitions in vascular smooth muscle cells (SMCs) in vitro. To determine the effects of arterial SMC-specific expression of a dominantnegative c-Myb molecule (Myb-Engrailed) on vascular remodeling in vivo, we performed carotid artery wiredenudation in 2 independent lines of binary transgenic mice with SM22␣ promoter-defined Doxycycline-suppressible expression of Myb-Engrailed. Adult mice with arterial SMC-speci… Show more

“…Furthermore, an immortalized mouse VSMC line would allow complementary in vitro studies of mouse models of cardiovascular disease in which, for example, transgenic manipulations and/or their effects have been restricted to VSMC in vivo (26,49). For these reasons, we developed and characterized a novel mouse VSMC cell line (MOVAS) and employed it in our study.…”

“…These mice are referred to as P4KO mice, with their wild-type littermates denoted as P4WT. Protocols for mouse tissue harvest and carotid artery wire denudation injury have been described (15) and were approved by the Animal Care Committee of University Health Network. Primary VSMC cultures were maintained in Dulbecco's modified Eagle's medium (DMEM) containing 10% fetal calf serum (Wisent; Montreal, Quebec, Canada), 50 ng/ml rat recombinant PDGF-BB (Sigma), and 1% penicillinstreptomycin in humidified air containing 5% CO 2 at 37°C.…”

“…24 Hearts from mice infected with group B coxsackieviruses were used as positive controls. 24 Cardiomyocyte cross-sectional area was measured at the level of the nucleus in 60 to 80 cells from each mouse with an imaging system 22 and Scion Image 4.0.1 software (Scion Corp). Longitudinal or oblique sections were excluded.…”
