Progesterone and oestradiol concentrations in the peripheral plasma of 12 dairy cows during the oestrous cycle were determined by competitive protein-binding and radioimmunoassay respectively. The levels of progesterone reached a peak of 6-7 ng/ml in mid-cycle, then began to fall 4 days before oestrus to values of about 1 ng/ml at oestrus and remained low for a further 4 days. Peak levels of oestradiol (11.2 ± 1.3 pg/ml, s.e.) occurred on the day before oestrus and a second peak (7.9 ± 2.9 pg/ml) occurred on Days 6-8 of the cycle.

Summary. Oestradiol was measured by radioimmunoassay in blood draining individual preovulatory follicles and in different ovarian tissues of the hen. The concentration of oestradiol in blood from follicles 20 to 50 hr before ovulation ranged from 66 to 264 pg/ml and was less than that in peripheral blood collected concurrently, suggesting a net uptake rather than secretion of oestradiol by the follicle at these times.In one bird approximately 6 hr before ovulation, the highest content of oestradiol in tissue (35 ng) was in the small (<5 mm) follicles and ovarian stroma; this represented 87\m=.\5% of the total ovarian content. Only 0\m=.\86 to 1\m=.\02ng oestradiol (2\m=.\0 to 2\m=.\5%) was found in the large preovulatory follicles and 2\m=.\22 ng (5\m=.\5%) in the postovulatory follicles. High concentrations of oestradiol (2\m=.\4ng/g) were also found in the liver.These results suggest that the small follicles and/or ovarian stroma are the main site ofoestradiol production in the fowl; they do not exclude the possibility that oestradiol is secreted at a high rate by the mature follicle for a short period immediately before ovulation, thus influencing the release of LH.

Summary. Testosterone, oestradiol and progesterone were measured in peripheral plasma during the oestrous cycle of 6 heifers. Oestradiol and progesterone results confirmed earlier reports. Concentration of testosterone on the day of oestrus was 40 \ m=+-\ 3 pg/ml (mean \ m=+-\ S.E.M.), and two peaks were detected during the cycle, one 7 days before oestrus (1809 \ m=+-\ 603 pg/ml) and the other (78 \ m=+-\ 7 pg/ml) on the day before the onset of oestrus. The concentration of progesterone declined in most cases 1 day after the maximum concentration of testosterone.Betamethasone treatment in 5 heifers extended luteal function by an average of 10 days: plasma androstenedione and oestradiol concentrations were unaltered; cortisol values were depressed for at least 16 days after treatment; testosterone concentrations were lowered by 13 \ m=+-\ 2\m=.\4%during treatment, and except in one heifer the peak on Day -7 was abolished.
