Concentration of Lipase from Aspergillus oryzae Expressing Fusarium heterosporum by Nanofiltration to Enhance Transesterification

Abstract: Nanofiltration membrane separation is an energy-saving technology that was used in this study to concentrate extracellular lipase and increase its total activity for biodiesel production. Lipase was produced by recombinant Aspergillus oryzae expressing Fusarium heterosporum lipase (FHL). A sulfonated polyethersulfone nanofiltration membrane, NTR-7410, with a molecular weight cut-off of 3 kDa was used for the separation, because recombinant lipase has a molecular weight of approximately 20 kDa, which differs fr… Show more

“…Additionally, they concluded that both enzyme-rich solutions can be efficiently reused (achieving FAME yields of around 80%-83%), and that the ceramic membranes tested did not exhibit any deterioration concerning their performance behavior. Similarly, Wijaya et al (2020) investigated the use of a sulfonated polyethersulfone nanofiltration membrane (3 kDa MWCO) for the condensation of an extracellular lipase expressed from A. oryzae. After nanofiltration, the concentrate stream presented an 83% increased lipase activity against the initial (unconcentrated) lipase solution.…”

“…Reinehr et al [23] adopted ultrafiltration and microfiltration to concentrate lipase produced from Aspergillus niger. Wijiya et al [24] used a sulfonated polyether sulfone nanofiltration membrane, with a molecular weight cut-off of 3 kDa, to concentrate lipase produced from recombinant A. oryzae. The total enzymatic activity increased at about fivefold after being separated by the membrane.…”

“…Response surface methodology was used to obtain a high concentration of the GABA strain under optimal fermentation conditions. There is also a study on nanofiltration membrane separation to concentrate lipase for biodiesel production [5]. The nanofiltration membrane technology developed showed a comparable fatty acid methyl ester (FAME) composition to that of commercial lipase, which improves its applicability and scalability.…”
