The testosterone levels with nasal gland volume and Leydig cell morphometry in sexually mature capybaras were compared using 10 adult capybaras, aging 28 months. Body and testicular weight were 50kg and 28g, respectively. The gonadosomatic index was 0.11±0.02%, while nasal gland volume was 34.12±13.04ml. Serum testosterone concentration was 25.07±14.73ng/ml. Leydig cell volume density and leydigosomatic index were, respectively, 28.8±8.3% and 0.030±0.007%.

Effects of Isotretinoin on the Reproduction of Pubertal Male Mice and Malformations in the Offspring

Isotretinoin is a vitamin A derivative, taken orally for the treatment of several dermatoses, including severe acne. This condition affects, principally, 80% of adolescents, males being the most affected. The objective of this study was to analyze the effects of isotretinoin on the reproductive system of pubertal males, and the possible teratogenic effect on the offspring. 1 mg/kg/day of isotretinoin was administered to pubertal male Swiss mice for 45 days, via gavage. On the 35th day the animals were placed to mate with females, after which they continued to be treated for another 10 days, and on the 46th day the male mice were euthanized. Vital and sexual organs of the males were analyzed for toxicity assessment and determination of reproductive function. On the 18th day of pregnancy the females were euthanized for evaluation of intrauterine development and the presence of congenital malformations in the offspring. The results demonstrated that the administration of isotretinoin did not promote toxicity in males, but decreased the weight of the reproductive organs, and decreased testosterone levels and the Johnsen score, including a decrease in Sertoli and Leydig cell numbers. Isotretinoin promoted alterations in the spermatic morphology of both head and tail, however, did not alter the fertility rate. Regarding the intrauterine development of the offspring, there was no impairment of fetal growth; however, the drug led to decreased fetal viability, increased resorption rate, and post implantation loss, added to which skeletal and visceral malformations were identified. It is concluded that isotretinoin presented a potential toxic effect on the male reproductive system by inducing spermatic alterations which had repercussions in offspring malformations.

RESUMOEstudou-se o efeito do Hormônio do Crescimento bovino (bGH) sobre os parâmetros seminais e a concentração sérica de testosterona de touros da raça Nelore. Dezesseis touros foram distribuídos em um delineamento fatorial 2 x 2 (duas idades e dois níveis de r-bST), com quatro animais em cada tratamento. As aplicações de rbST foram realizadas a cada 14 dias, totalizando nove aplicações por animal, em um período experimental de 120 dias. As coletas de sêmen foram realizadas a cada 15 dias e, para determinar as concentrações de testosterona, foram realizadas coletas de sangue a cada quatro horas, num total de 24 horas, e a cada 30 dias a partir da primeira aplicação de r-bST. O sêmen dos touros adultos tratados com r-bST apresentaram maior motilidade e vigor (P<0,05) em relação aos animais controles (79,70±11,56% vs 64,06±23,65%) e (3,80±0,88 vs 2,92±1,25), respectivamente; entretanto, as demais características seminais não foram influenciadas (P>0,05) pelo tratamento com r-bST. Os aspectos físicos do sêmen dos touros jovens não foram afetados (P>0,05) pelo tratamento com r-bST. Entretanto o tratamento com r-bST diminuiu os defeitos morfológicos menores (delgado, gigante normal, pequeno normal, globoso, cabeça isolada normal, abaxial, cauda dobrada e enrolada levemente e gota distal) em relação aos jovens não tratados (P<0,05). As concentrações séricas de testosterona foram influenciadas (P<0,05) pelo tratamento, aos 120 dias após a primeira aplicação do r-bST, e apresentaram tendência de três picos no período de 24 horas.Palavras-chave: touro, hormônio, sêmen, Somatotrofina bovina recombinante (r-bST), testosterona MOT and VIG (P<0.05) ABSTRACT The aim of this trial was to study the effect of Growth Hormone (GH) on the seminal parameters and testosterone profile from Nellore bulls. Sixteen bulls were allocated in a 2 x 2 factorial arrangement (two ages and two r-bST levels), with four animals per treatment. The r-bST was injected every 14 days, with a total of nine injections per animal, during the 120 days of the experimental period.
