Concanavalin A Derivatives with Altered Biological Activities

Abstract: Chemical derivatization of tetrameric concanavalin A (Con A) with succinic anhydride or acetic anhydride converts the protein to a dimeric molecule without altering its carbohydrate-binding specificity. At low concentrations, the dose-response curves for the mitogenic stimulation of mouse spleen cells by native Con A and succinyl-Con A are similar. Above lectin concentrations of 10 ug/ml, however, the response to Con A is diminished, while that for succinyl-Con A does not decrease until much higher doses are r… Show more

“…Since the identical dose-response curves were unexpected , we carried out hemagglutination and tnitogenicity tests to assure the authenticity of our succiny l Con A prepar ation ; in both assays the results agreed well with literature values [6].…”

“…Con A has been shown to inhibit patching and capp ing of various surface receptors on lymphocytes and numerous established cell lines [4, 141 . This inhibition is reversed by microtubule depolymerizing agents such as coichicine [7] and canno t be elicited b y succiny l. Con A [4,6], a monovalent derivative of Con A . As assayed in our system , Con A and succi nyl Con A are eq ua ll y effective at altering the 37°C binding pat tern.…”

“…The decreased thymidine uptake observed at high lectin concentrations appears to correlate with impaired lymphocyte survival (Fig. 7 B), perhaps a result of the inactivation of surface receptors necessary for membrane function (23). Little correlation was observed in Con A-stimulated lymphocyte cultures between DNA synthesis and MAF production; these may constitute two independent phenomena triggered simultaneously by lectins.…”

“…Effect of Chemical modification on haemagglutinating activity of the field bean or lablab bean lectin. On the other hand acetylation, and succinylation of Con A did not alter its lectin activity as well as its antigenicity (17,18). Citraconylation of potato lectin also did not alter its biological activity (19).…”
