Computational models applied to metabolomics data hints at the relevance of glutamine metabolism in breast cancer

Abstract: Background: Metabolomics has a great potential in the development of new biomarkers in cancer and it has experiment recent technical advances. Methods: In this study, metabolomics and gene expression data from 67 localized (stage I to IIIB) breast cancer tumor samples were analyzed, using (1) probabilistic graphical models to define associations using quantitative data without other a priori information; and (2) Flux Balance Analysis and flux activities to characterize differences in metabolic pathways. “… 12 14 Previous studies demonstrated significant differences in glutamine and alanine metabolism between ER-positive and ER-negative breast cancers. 12 , 13 Besides, the MYC -driven accumulation of 2-hydroxyglutarate has been shown to promote breast cancer progression. 14 However, large-scale metabolomic researches connecting metabolism to TNBC genomics are still lacking.…”

“…Tumor cells convert 90% of glucose and 66% of glutamine into lactate and alanine. Glutamine and glutamate contribute to the carbon backbone in the TCA cycle, and this is relevant in conditions of carbon diversion to glycolysis [192,193]. The transfer of an amino group from glutamate to oxaloacetate via aspartate aminotransferase results in α-ketoglutarate and aspartate, whereas nitrogen transfer from glutamate to pyruvate via alanine aminotransferase results in α-ketoglutarate and alanine.…”

“… 9 It mainly investigated the quantities, types and changes of endogenous metabolites in biological systems such as blood, urine, and tissues by the application of newly developed analytical methods. 10 Metabolomics is now widely used in the diagnosis of disease 11 , 12 and biomarker screening. 13 , 14 Nuclear magnetic resonance (NMR) is a kind of analytical methods which is a noninvasive technique 15 and an invaluable tool for determining the structure, composition and function of complex biological molecules.…”
