Computational modelling of γ-H2AX foci formation in human cells induced by alpha particle exposure

Abstract: In cellular experiments, radiation-induced DNA damage can be quantified by counting the number of γ-H2AX foci in cell nucleus by using an immunofluorescence microscope. Quantification of DNA damage carries uncertainty, not only due to lack of full understanding the biological processes but also limitations in measurement techniques. The causes of limited certainty include the possibility of expressing foci in varying sizes responding individual DSBs and the overlapping of foci on the two-dimensional (2D) immun… Show more

“…6) where one could observe that gamma-H2AX foci per cell follows a linear trend from 0 to 10 cGy beyond which the curve bends downward between 10 and 50 cGy and resumes the linear trend in the range 50 to 500 cGy. The overlapping of the gamma-H2AX foci at doses of 1 Gy induced by alpha radiation was observed by Abu Shqair et al [25].…”

“…In doing so, a model function was applied that explicitly accounted for the clustering of ion tracks which lead to indistinguishable foci. In this way, the resulting effect on the time evolution of observed foci is included as an important ingredient that goes beyond models estimating miscounting due to the overlapping of foci [22,23].…”

“…Currently, chemical effects in BP can address many topics in fields of primary concern: radiation protection and dosimetry 20 , space radiation protection 6 , 21 , alpha rays in nuclear industry 22 , alpha and Boron neutron capture therapy (BNCT) 23 , 24 . To take advantage or to protect from damage occurring in BP, understanding the radiation chemistry in this region with a high spatial resolution is necessary.…”
