Comprehensive and quantitative profiling of lipid species in human milk, cow milk and a phospholipid‐enriched milk formula by GC and MS/MSALL

Abstract: Here we present a workflow for in-depth analysis of milk lipids that combines gas chromatography (GC) for fatty acid (FA) profiling and a shotgun lipidomics routine termed MS/MSALL for structural characterization of molecular lipid species. To evaluate the performance of the workflow we performed a comparative lipid analysis of human milk, cow milk, and Lacprodan® PL-20, a phospholipid-enriched milk protein concentrate for infant formula. The GC analysis showed that human milk and Lacprodan have a similar FA p… Show more

“…The monounsaturated FA (MUFA) (23%) content was predominantly oleic acid (C18:1c9, 17%), whereas polyunsaturated FA (PUFA) was low (~1% on total FAME) and mainly accounted for by linoleic acid (LA, C18:2 c9, c12). These results are similar to those reported in previous studies in a commercial BM samples (Sokol et al [18]; Kristensen et al [19]).…”

“…11,12 In such studies ions are sequentially separated by an initial mass analyzer followed by collisional activation with measurement of all product ions for that mass. This process is repeated for the entire mass range of interest.…”
