Comprehensive analysis of nine monoamines and metabolites in small amounts of peripheral murine (C57Bl/6 J) tissues

Abstract: Monoamines, acting as hormones and neurotransmitters, play a critical role in multiple physiological processes ranging from cognitive function and mood to sympathetic nervous system activity, fight-or-flight response and glucose homeostasis. In addition to brain and blood, monoamines are abundant in several tissues, and dysfunction in their synthesis or signaling is associated with various pathological conditions. It was our goal to develop a method to detect these compounds in peripheral murine tissues. In th… Show more

“…Plasma insulin levels were analysed using a commercially available ELISA kit (Alpco Diagnostics, Salem, NH, USA) following the manufacturers' instructions. Plasma catecholamines were analysed and detected with an HPLC system coupled with an electrochemical detector (EcD) as described previously [14]. The sample clean-up was performed according to the protocol described by Recipe (Recipe, Munich, Germany).…”

“…Therefore, diseases such as hypertension and myocardial infarction can be triggered when the content of NE is abnormal in the body . In recent years, the studies of NE have received more and more attention of researchers . Meanwhile, a variety of analysis methods have been reported to detect NE, which including high‐performance liquid chromatography, electrochemical detection, capillary electrophoresis, and chemiluminescence .…”

“…In recent years, the studies of NE have received more and more attention of researchers . Meanwhile, a variety of analysis methods have been reported to detect NE, which including high‐performance liquid chromatography, electrochemical detection, capillary electrophoresis, and chemiluminescence . However, these analysis methods always have the disadvantages such as time‐consuming and complex during the operation.…”

“…Because the concentration of catecholamine in tissues declined exponentially, we could obtain the rate constant of NE efflux (expressed in h-1). Comprehensive analysis of norepinephrine was carried out by reverse-phase liquid chromatography with electrochemical detection as described by 69 . The values obtained were expressed as ng/mg wet tissue and were logarithmically transformed for calculation of linearity of regression, standard error of the regression coefficients and significance of differences between regression coefficients.…”
