Complex Regulation by Apetala2 Domain-Containing Transcription Factors Revealed through Analysis of the Stress-Responsive TdCor410b Promoter from Durum Wheat

Abstract: Expression of the wheat dehydrin gene Cor410b is induced several fold above its non-stressed levels upon exposure to stresses such as cold, drought and wounding. Deletion analysis of the TdCor410b promoter revealed a single functional C-repeat (CRT) element. Seven transcription factors (TFs) were shown to bind to this CRT element using yeast one-hybrid screens of wheat and barley cDNA libraries, of which only one belonged to the DREB class of TFs. The remaining six encoded ethylene response factors (ERFs) belo… Show more

“…It is known that ERFs specifically bind to the GCC‐box and sometimes to CRT DNA cis ‐elements, whereas DREB TFs specifically recognize DRE and CRT sequences (Eini et al, ). The affinities of the GCC‐Box, DRE, and CRT cis ‐elements to TaSHN1 were determined using an Y1H assay (Figure c).…”

“…With the aim to decrease differences in the developmental phenotypes of control and transgenic wheat plants, a stress‐inducible promoter of barley, Dehydrin 8 ( Dhn8 ), was also used for overexpression of TaRAP2.1L . The structure and properties of the Dnh8 promoter (James et al ., ; Xiao and Xue, ) are similar to those of the TdCor410b promoter (Eini et al ., ), showing strong induction by cold, drought and salinity, a moderate level of constitutive expression in leaves and a high level of expression in the developing grain. A total of 12 independent transgenic lines of wheat (cv.…”

“…Members of the ERF subfamily differ from DREBs/CBFs in their recognition of specific cis ‐elements (Canella et al ., ; Eini et al ., ; Hao et al ., ; Sakuma et al ., ). Most studied ERFs recognize and serve promoters containing GCC‐box elements (AGCCGCC), although in some cases, they can either additionally or solely bind to CRT elements (GCCGAC) (Eini et al ., ). While the mechanisms for positive regulation of downstream stress‐related genes by ERFs and DREB transcription factors (TFs) are reasonably well studied, mechanisms of negative regulation by the members of the DREB and ERF subfamilies, containing repressor motifs, are less well understood.…”

“…The generation of vectors for plant transformation has been described previously (Eini et al ., ; Kovalchuk et al ., ; Morran et al ., ). Briefly, the 2x35S promoter was excised from the pMDC32 vector (Curtis and Grossniklaus, ) and replaced with one of three promoters: the constitutive ZmUbiquitin promoter (Eini et al ., ), or stress‐inducible OsWRKY71 or TdCor39 promoters (Kovalchuk et al ., ), resulting in pUbi, pWRKY71 and pCor39 vectors, respectively. A 767‐bp fragment of the TaHDZipI‐5 coding sequence (CDS) (GenBank accession ) (Harris et al ., ) was isolated from T. aestivum L. cv.…”
