The cytogenic origin of canine transmissible venereal tumor (CTVT) still remains unknown. Resulting from paucity of information on the histiocytic phenotypic features and behaviours of CTVT, this study was undertaken to show some uncommon cytomorphological features of CTVT and its immunoreactivity with S-100 protein. Nine cases of CTVT were investigated (6 females and 3 males) using nine fine needle aspirates for cytology and four biopsy samples for histopathology and immunohistochemistry. Cytology revealed CTVT cells with pale basophilic, fine granular cytoplasm and distinct intra-nuclear (3/9) and intra-cytoplasmic (9/9) vacuolations. Erythrophagocytosis by a giant binucleated CTVT cell in one dog and nuclear budding in 2 dogs were observed. Histopathology showed loose sheets and cords of uniformly round to ovoid cells with slight indistinct eosinophilic cytoplasm and a tendency towards glandular as well as syncytial formation. Nuclei were large and round with a single centrally placed nucleolus. The mitotic index was high. Immunohistochemistry of the four biopsy samples revealed negative immunoreactivity to pan-cytokeratin, actin and desmin but positive immunoreactivity with vimetin and S-100 proteins. This is the first report in which CTVT showed erythrophagocytosis, nuclear budding and positive immunoreactivity to S-100 protein. Based on these cytomorphological and immunohistochemical features, we conclude that CTVT is of histiocytic/dendritic origin.

An eight-month old female entire mixed breed dog in Arizona was presented for a 15-day history of multifocal cutaneous nodules and plaques, with progressive stertor, anorexia, haemorrhagic gingival masses, peripheral lymphadenopathy and mild haemorrhagic vaginal discharge without obvious genital masses. Fine needle aspirate cytology was consistent with multifocal cutaneous and mucosal canine transmissible venereal tumour (TVT) with peripheral lymph node metastases. Histopathology and immunohistochemistry supported the diagnosis. Eight weekly treatments with 0.5 mg/m2 intravenous vincristine resulted in complete clinical resolution, without relapse five months after treatment. The classic presentation of TVT is of adult dogs with genital tumours and low metastatic rate. This report describes an unusual presentation in a puppy with multifocal, extragenital, metastatic, cutaneous and mucosal TVT. In doing so, it underscores the importance of recognising less common presentations of TVT, as well as diagnostic cytological and histopathological findings, as prognosis for cure is excellent with treatment.

Transmissible venereal tumor (TVT) is a neoplasm that can be transmitted between female and male dogs via mating by the physical transfer of viable tumor cells. The tumor may spread to different parts of the body through sniffing, licking, scratching or biting affected lesions. It can be also observed in wild carnivores. TVT is mostly located in the posterior wall of the vagina, in the joint of vaginal vestibule and vagina in female dogs, while it is usually located in caudal part of the penis in males. Initially, the tumor is small and varying from pink to red, subsequently progressing to a large, ulcerated, and contaminated mass. By the time, the tumor's volume increases and the lesions become friable, hyperemic, hemorrhagic, multilobular and cauliflower-like masses. A serosanguinous vaginal discharge is observed. Definitive diagnosis is based on anamnesis, location of the mass, cytological and histological findings. The cells in the slides vary from round to oval or variable shapes and contain a pale blue or colorless cytoplasm with a single distinctive nucleus. They include small, light, clear intracytoplasmic vacuoles. TVT should be immediately treated as soon as diagnosed because there is no method to prevent it. Chemotherapy is the effective method for the treatment of TVT with the intravenous use of vincristine sulfate, which is a chemotherapeutic agent, once in a week for approximately three weeks applications. The cure rate varies at 90-95%. In this review, the incidence, etiopathogenesis, clinical findings, diagnosis, prognosis and therapies of TVT have been emphasized. Transmisibil Venereal Tümör: Derleme ÖZTransmisibil Venereal Tümör (TVT), erkek ve dişi köpeklerin çiftleşme sırasında canlı tümör hücrelerinin fiziksel transferi ile bulaşabilen neoplastik bir yapıdır. Tümör etkilenen lezyonların koklanması, yalanması, tırmalanması ve ısırılmasıyla vücudun farklı bölgelerine yayılabilir. TVT aynı zamanda vahşi kornivorlarda da gözlenebilir. Tümör genellikle erkek köpeklerde penisin kaudal kısmında lokalize olurken, dişilerde çoğunlukla vajinanın posteriyör bölgesinde, vestibulum ve vajinanın birleşme yerinde şekillenir. İlk başlarda tümör küçük ve pembeden kırmızı renge değişen şekildedir, daha sonra büyük ülserasyonlu ve kontamine bir kitle haline dönüşür. İlerleyen zamanlarda tümörün hacmi artar ve şekillenen lezyonlar multilobüler, karnıbahar görünümünde olup gevrek, hiperemik ve hemorajik yapıdadırlar. Serösangionöz vajinal akıntı gözlenir. Tümörün kesin tanısı anemnez bilgilerine, kitlenin oluşum yerine, sitolojik ve histolojik bulgulara dayanmaktadır. Preparatlardaki hücreler yuvarlak, oval veya değişik yüzeyli yapıda olup, solgun mavi veya renksiz sitoplazma ile belirgin bir nükleus içerirler. Hücrelerde küçük, parlak ve temiz görünümlü intrastoplazmik vakuoller bulunur. TVT'nin oluşumunu tamamen engelleyecek bir yöntem olmadığı için, teşhis edildiğinde derhal tedavi edilmesi gerekir.
