Complete nucleotide sequence of Colorado tick fever virus segments M6, S1 and S2.

Abstract: The nucleotide sequences of the tenth (M6), eleventh (S1) and twelfth (S2) dsRNA genomic segments of the Florio strain (N-7180) of Colorado tick fever virus were determined and found to be 675, 998 and 1884 bp, respectively, in length. A nonanucleotide motif and a hexanucleotide motif were found to be highly conserved in the 5h and 3h non-coding regions (NCRs), respectively, of the three segments. The first and last three nucleotides of each segment were of inverted complementarity, and segment-specific invert… Show more

“…Bars in each histogram labeled with the same letters are not significantly different (P Ͼ 0.05) according to the least-significant difference test. tion (44,45). BdCV1 dsRNAs contain the sequence CGCAAAAA AGAAGAAAAG at the 5= termini, and this sequence is similar to that (GCAAAAAAGAGAAUAAAG) of MoCV1, a close tentative member of the genus Chrysovirus (28), and to that (GAUAAAA AAA) of some other members of this genus, e.g., Aspergillus fumigatus chrysovirus (AfuCV) (46), PcV (27), and Helminothosporium victoriae 145S virus (HvV145S) (47).…”

“…BHK-21 cells were cultured as monolayers or suspensions as previously described (Attoui et al, 1997). C6\36 mosquito cells were cultured as monolayers in Leibovitz's L-15 medium supplemented with 5 % FBS, 2 % tryptose phosphate broth, penicillin G (100 IU\ml) and streptomycin (100 µg\ml).…”

“…Other members of the genus are Eyach virus, isolated in Germany (Rehse- Ku$ pper et al, 1976), and two Eyach virus antigenic variants (AR577 and AR578), isolated in France (Chastel et al, 1984). In previous studies, an antigenic relationship between these American and European isolates was established (Chastel et al, 1984 ;Karabatsos et al, 1987) and full-length nucleotide sequences of genomic segments 10, 11 and 12 of CTF virus were determined (Attoui et al, 1997). Other viruses are also considered as tentative species of the genus, e.g.…”

“…Until the identification of the genomic sequences of CTF virus, the diagnosis of viral infection was made by virus isolation and characterization on one hand, and by ELISA on the other. 15 Recently, we sequenced the M6, S1, and S2 genomic segments 19 of CTF virus (GenBank accession numbers AF000720, U72694, and U53227, respectively), as well as a part of the M5 genomic segment (GenBank accession number AF007172) of the Florio strain of CTF virus. This led to the development of molecular diagnostic procedures based on the S2 genomic sequence.…”

“…15 Moreover, many strains were reported 16,17 that could be detected by RNA-RNA blot hybridization. 18 Recently, we determined the first genomic sequences of the M6, S1, and S2 ds RNA segments of the Florio strain of the CTF virus 19 (GenBank Accession numbers AF000720, U72694, and U53227, respectively). In this paper, we report molecular and serologic procedures for the diagnosis of infection by both American and European isolates of Coltiviruses.…”
