Complete nucleotide sequence of a putative new cytorhabdovirus infecting lettuce

Abstract: The full-length nucleotide sequence of the genomic RNA of a new cytorhabdovirus infecting lettuce was determined. Six open reading frames were found in the antigenomic sequence of the 12,926-nt negative-sense viral RNA genome. The genomic organisation was similar to that of lettuce necrotic yellows virus (LNYV), the type member of the genus Cytorhabdovirus: 3'-N-P-3-M-G-L-5', where N is the capsid protein gene, P the putative phosphoprotein gene, 3 a gene coding for a putative protein of unknown function, M th… Show more

“…Furthermore, a transmembrane domain was identified in the C-terminal region (aa positions 508-530) ( Table 1). Similar domains were also reported in this region for other plant rhabdoviruses (Bandyopadhyay et al, 2010;Dietzgen et al, 2006;Heim et al, 2008).…”

“…The amino terminal region of the ADV G protein is hydrophobic, and its overall hydrophilicity plot (data not shown) is similar to those of the G proteins of LYMoV (Heim et al, 2008) and lettuce necrotic yellows virus (LNYV) (Dietzgen et al, 2006). In ADV G, a signal peptide consisting of 30 aa and containing a IGD¼ R (¼indicates the predicted cleavage site) was identified.…”

“…The lettuce necrotic yellows virus (LNYV), identified in 1963 in Australia (4), is found only in Australia and New Zealand, but it is very similar to the lettuce yellow mottle virus that was recently isolated in France (5). Infection by LNYV causes fading of the leaves and necrosis of the heads, rendering the lettuce unsuitable for consumption.…”

“…However, to date all the fully sequenced plant rhabdoviruses have an additional open reading frame between the P and M genes (Jackson et al 2005;Bourhy et al 2005;Heim et al 2008). There are six blocks of known amino acid sequence conservation in the L protein of the rhabdoviruses.…”
