Complete Genome Sequence of Klebsiella pneumoniae 1084, a Hypermucoviscosity-Negative K1 Clinical Strain

Abstract: fWe report the complete genome sequence of Klebsiella pneumoniae 1084, a hypermucoviscosity-negative K1 clinical strain. Sequencing and annotation revealed a 5,386,705-bp circular chromosome (57.4% G؉C content), which contains 4,962 proteincoding genes, 80 tRNA genes, and 25 rRNA genes.

“…Up to 99% of the reads (13.1 to 36.6 million reads per isolate with a median of 20 million reads; http://onlinelibrary.wiley.com/doi/10.1111/1462-2920.13016/suppinfo) could be mapped to a non‐redundant gene list of the K. pneumoniae pan‐genome. This pan‐genome was generated on the basis that the fully sequenced genome of the clinical strain 1084 (sequence type 23) (Lin et al ., ) complemented with publicly available DNA sequence data of 10 K. pneumoniae isolates (see Materials and methods for details) to gain comprehensive and unbiased insights into the transcriptional landscape of this pathogen. To exclude any bias in mapping efficiency due to the choice of the core genome reference strain, we also created a pan‐genome based on a different sequence type (MGH 78578; ST38).…”

“…A total of 30 sRNAs were included in the final gene set. To generate a single reference sequence file, sequences of core‐genom genes were selected from the sequence type 23 strain ‘1084’ (Lin et al ., ), and the accessory genes and the singletons were added from each of the remaining 10 genomes considered in this study. The origin of each sequence is indicated by the locus ID in http://onlinelibrary.wiley.com/doi/10.1111/1462-2920.13016/suppinfo (see also Table ).…”
