Complete Genome Sequence Analysis of a Reassortant Strain of Bluetongue Virus Serotype 16 from Italy

Abstract: The complete genome sequence of a reassortant field strain of bluetongue virus serotype 16 (BTV-16), isolated from cattle in the Apulia region of Italy in 2002, has been determined by Illumina sequencing. Sequence comparisons of segment 1 (Seg-1) to Seg-10, except Seg-5, show that BTV-16 strain ITL2002 belongs to the major eastern topotype of BTV.

“…Despite the absence of any vaccination against this serotype and specific movement control policies, BTV‐16 virus circulation was quite limited in few geographical areas of Italy. The reasons behind the limited spread of this serotype are not fully clear (Lorusso et al., ).…”

“…Information based on partial genome sequences from a limited numbers of BTV field strains, indicated that reassortment between some strains had occurred (e.g. for BTV-16(e) in Italy 2002 or BTV-1(w) and BTV-4(w) in Sardinia 2012 [ 38 40 ]).…”

“…As for Seg‐6, 11 putative nucleotypes (A–K) are potentially designated but its sequence variation does not show an absolute correlation with serotype (Maan et al., ; Schulz et al., ; Sun et al., ). Importantly, a characteristic of BTV is the ability to “reassort” genomic segments with related members of the same species (Lorusso, Costessi et al., ; Lorusso, Sghaier et al., ; Nomikou et al., ). BTV strains derived from modified live‐attenuated vaccines (MLVs) have also been used for BT prevention and isolated in the field (Batten, Maan, Shaw, Maan, & Mertens, ; Ferrari et al., ; Savini et al., ).…”
