Complete amino acid sequence of human placental protein 14: a progesterone-regulated uterine protein homologous to beta-lactoglobulins.

Abstract: Placental protein 14 (PP14), also known as progestagen-dependent endometrial protein and pregnancyassociated endometrial a2-globulin, is synthesized by the human secretory endometrium and decidua. We have isolated from a human decidual cDNA library clones corresponding to PP14 and deduced its entire amino acid sequence. PP14 contains 180 amino acids, 18 of which correspond to a putative signal peptide. The predicted molecular weight of the pre-PP14 is 20,555 and that of the mature protein is 18,787. PP14 is en… Show more

“…The HZI decreased in a dose-dependent manner as the concentration of 125 I-ZIF-1 or cold ZIF-1 increased. There is a discrepancy in the efficiency of spermatozoa zona pellucida binding inhibition of glycodelin-A between this report and two previous studies (5,11). Several possibilities might account for this.…”

“…The major protein product expressed between the late luteal phase of the menstrual cycle and the first trimester of pregnancy is a progesterone-regulated lipocalin called PP14 [89,90] ; although first isolated from placenta, its principal site of synthesis is the secretory glandular epithelium of the endometrium and the decidua [91]. PP14 is also present in high concentrations in amniotic fluid, uterine luminal fluid, and at lower levels in plasma, but is not expressed by other tissues [89].…”

“…analysis of CNBr-digested glycodelin-S was performed using an on-line microbore reverse-phase high performance liquid chromatography system (Brownlee C 18 Aquapore column) coupled to a VG BioQ triple quadrupole electrospray mass spectrometer. The sample was dissolved in 0.1% trifluoroacetic acid (solvent system A) for injection on the column.…”

“…To address this anomaly, we have initiated comparative structure/function studies of GdA and GdS. GdA, whose protein sequence has been determined by cDNA sequencing (18), is a 162-amino acid glycoprotein that gives a 28-kDa band upon SDS gel electrophoresis (16). GdS shows a major band at 27 kDa and a minor band at 30 kDa when analyzed in the same manner (19).…”
