Competitive Upconversion-Linked Immunosorbent Assay for the Sensitive Detection of Diclofenac

Abstract: Photon-upconverting nanoparticles (UCNPs) emit light of shorter wavelength under near-infrared excitation and thus avoid optical background interference. We have exploited this unique photophysical feature to establish a sensitive competitive immunoassay for the detection of the pharmaceutical micropollutant diclofenac (DCF) in water. The so-called upconversion-linked immunosorbent assay (ULISA) was critically dependent on the design of the upconversion luminescent detection label. Silica-coated UCNPs (50 nm i… Show more

“…of the blank measurements) and p is the slope of the calibration curve. The factor k = 3 was used in compliance with the IUPAC rules (Inczedy et al, 1998;IUPAC, 2014). The time-dependent increase in the optical signal in the presence of diclofenac was fitted using the modified Gompertz model (Zwietering et al, 1990):…”

“…Bioimaging probes are usually composed of organic fluorophores or fluorescent proteins, but they produce strong background fluorescence in the visible light region and have poor light stability, which greatly limits their biomedical applications [ 135 137 ]. Quantum dots (QDs) have high photostability, it is difficult to make extensive use of them in vivo due to their toxicity and side effects.…”

“…Based on the same principle, we can see the signal from FNPs that are attached to the fiber through linkage between the spike protein and antispike protein antibody tagged with FNPs conjugated with antihuman antibodies. Similar assays have been used to detect water pollutants [36].…”
