Competition between cancer cells and T cells under immunotherapy: a structured population approach

Abstract: Abstract. T cells are key players in the immune action against the invasion of cancer cells. During an immune response, antigen-specific T cells dynamically sculpt the antigenic distribution of cancer cells, and cancer cells concurrently shape the repertoire of antigen-specific T cells. The succession of these reciprocal selective sweeps can result in "chase-and-escape" dynamics, and lead to immune evasion. It has been proposed that immune evasion can be countered by immunotherapy strategies aimed at regulatin… Show more

“…According to the World Health Organization (WHO), in 2020, the last year of available data, around 1.93 × 10 7 new cases were registered, having an estimated mortality of almost 1 × 10 7 , constituting approximately 1∕6 of total human deaths in the world [1]. Among the treatments used to fight cancer, there are those based on purely medical (surgery [2]), physical (radiotherapy [3][4][5][6][7]), chemical (chemotherapy [8][9][10][11]), biological (immunotherapy [12,13], targeted gene therapy [14], oncolytic [15] and hormonal) aspects, and combinations of the above mentioned. Cancer has acquired singular importance in recent decades due to its impact on society and the efforts dedicated to combating it.…”

“…Despite this, only a small reduction in either the probability of a cytotoxic effector cell killing or deactivating a tumor cell, or the rate of influx of these lymphocytes into the tumor vicinity, has been found sufficient to provoke tumor regrowth [86]. It is clear that overcoming lapses in immune response which can lead to the tendency for regrowth is crucial: immune memory boosting in combination with other treatment protocols has demonstrated the potential to help to establish stable dormancy [88].…”
