Comparison of Water Displacement and WinRHIZO Software for Plant Root Parameter Assessment

Abstract: Water-displacement and WinRHIZO root-scanning methods were compared for efficacy of root damage assessment. Results from both methods were similar and a highly significant relationship was found between the two methods in trial one (r2 = 0.9968, P < 0.0001) and trial two (r2 = 0.9988, P < 0.0001). Both protocols provide consistent root volume measurements; however, water displacement is preferred as an economical method if a quick evaluation of a large amount of roots is essential. For a more detailed ro… Show more

“…The root morphological characteristics analysis was conducted using a WinRHIZOPro analysis system (Regent Instruments, Quebec, QC, Canada) (Bouma et al, 2000). However, root volume was evaluated with water displacement technique due to large quantity of roots (Pang et al, 2011). Biomass of leaf, root, stem and root nodule was estimated by drying in a hot air oven at 75°C until a constant weight was obtained.…”

“…WinRHIZOPro is an image analysis system for automatic root measurement in morphology (that is, root length, root surface area, root tip numbers, etc.). Total root volume was measured with water displacement method (Pang et al, 2011). Roots and stems were then dried in an oven at 75°C till a constant weight.…”

“…For each plant, root morphological traits, that is, taproot length, total root length, external root surface root area and root tip number were measured with the WinRHIZOPro Image Analysis System (Regent Instruments, Quebec, Canada) (Bouma et al, 2000). Water displacement method was used for measuring total root volume (Pang et al, 2011). Then, roots and stems were dried in an oven at 75°C till constant weight for biomass measurement.…”
