Comparison of two long‐acting preparations of metoprolol with conventional metoprolol and atenolol in healthy men during chronic dosing.

Abstract: 1 Eight healthy men received two long-acting formulations of metoprolol 200 mg (SA Astra, SR Geigy), conventional metoprolol 200 mg and atenolol 100 mg once daily for 1 week each in balanced, crossover fashion. There was a washout period of at least a week between each phase. 2 On the last day of each phase, post-exercise heart rate was recorded at intervals and compared to pretreatment values. Plasma metoprolol concentrations were measured. 3 The mean AUC was similar after each of the three formulations of me… Show more

“…Exercise testing was carried out as described by Freestone et al (1982) Eighty six EMs (50 males, 36 females; mean age 51 years, range 22-73 years) and 11 PMs (six males, five females; mean age 47 years, range 27-62 years) took part in the study. significantly less of both the hydroxy-and H117-04 metabolites ( the M/H117-04 ratios were 0.71 (P < 0.001) and 0.46 (P < 0.001), respectively.…”

“…In EM the mean elimination halflife was similar to that reported by others (2.8 h) but it was much longer in PM (7.6 h). (Freestone et al, 1982).…”

“…'long-acting' formulations of metoprolol was associated with marked variability in 8-adrenoceptor blockade 24 h after a 200 mg dose, and with insignificant f3-adrenoceptor blockade in the most extensive metabolisers (metabolic ratio < 1) (Freestone et al, 1982). Therefore, these formulations may offer no advantage over the conventional tablets since they too must be prescribed twice daily in angina unless the oxidation phenotype is known.…”

“…Its short elimination half-life has led to the development of a slow-release formulation of the polymer-matrix type (Lopresor SR®). Clinical studies have shown that this formulation is effective and well tolerated when administered once daily (Nievel & Harvard, 1981) but its ability to sustain statistically significant 3-adrenoceptor blockade throughout 24 h has recently been questioned (Freestone et al, 1982).…”
