Comparison of the Stimulating Capacity of Human Leukemia T-Cell, Null Cell, and B-Cell Lines in Mixed Lymphocyte Culture: Brief Communication

Abstract: One-way mixed lymphocyte reactions between lymphocytes from normal human donors and mitomycin C-treated cells from human leukemia T-cell, null cell, and B-cell lines were investigated. An Epstein-Barr virus (EBV)-negative Japanese Burkitt's lymphoma line and two EBV-positive normal lymphoid cell lines were studied in parallei. Normal lymphocytes were stimulated significantly by the cultured null cells and B-cells, but only slightly by the cultured T-cells. The stimulatory capacity of these two leukemia cell li… Show more

“…In this and other studies, all tested non-Bhon-T-lymphoid malignancies also expressed these serologically determined "Ialike" determinants , Greaves & Janossy 1978, Greaves 1981. Because of the close association of MLC stimulatory determinants with HLA-DR antigens, the lack of stimulatory capacity of T-cell tumors is not unexpected and was in fact observed earlier with fresh tumor cells and T-cell leukemia-derived cell lines , Leventhal et al 1977, Han & Minowada, 1978, Han, Dadey & Minowada, 1979, Isuboto et al 1978.…”

“…MLTR has been used to analyse the cellular antigens on tumor cells, showing the in vitro proliferation of responding lymphocytes by the stimulation of tumor cells (Tsubota et al 1978;Grimm et al 1984). Known candidates of epitopes which evoke MLTR are histoincompatible Ia and tumor specific antigens (Reinsmoen et al 1978; Han et al 1979).…”

“…Spontaneous rosette formation with sheep erythrocytes (EN), receptors for complements, C3b (human serum, EAChu) and C3d (mouse serum, EACm°), and Fe recepotors for IgG (EAIgG) and IgM (EAIgM) were examined as described previously (Tsuchiya et al 1980a). Cell surface immunoglobulins (sIg) and cytoplasmic immunoglobulin (cIg) were detected by direct immunofluorescence.…”

“…-Ia-like antigen ; null cell line ; mixed lymphocyte reaction There has been a number of reports about in vitro stimulation of leukemia cells in autologous or HLA-identical (allogeneic) mixed lymphocyte culture (MLR) (Fefer et al 1974(Fefer et al , 1976. The stimulating capacities of established leukemia cell lines on allogeneic MLR have also been demonstrated (Han et al 1977;Tsubota et al 1978). Multiple factors may be involved in autologous MLR with leukemia cells ; immunocompetence of patients during chemotherapy (Hersh and Oppenheim 1967;Gutterman et al 1972), and autologous stimulation of normal lymphocyte subpopulations (Opelz et al 1975).…”
