Comparison of the specificity of human and bovine tuberculin PPF for testing cattle. 3. National trial in Great Britain

Abstract: A field trial on a country-wide basis was undertaken to compare the specificity for bovine tuberculosis of single and comparative tuberculin tests in cattle using either Weybridge human or Weybridge bovine PPD. The tests were made on 10,305 cattle in 179 herds distributed throughout all regions of England, Scotland and Wales. Results showed that a comparative tuberculin test using avian PPD with either human or bovine PPD had a much higher efficiency than a single injection of mammalian tuberculin in the neck… Show more

“…High specificities for the skin tests are also described in reviews [13][14][15], all citing a paper from Lesslie [33] published in 1975. The tuberculins used at that time might differ from those used nowadays due to efforts in standardization.…”

“…The influence of environmental mycobacteria and non-specific responses on TST in the diagnosis of bovine TB have been widely reported (Philips et al 2003;Biet et al 2005;de la Rua-Domenech et al 2006a;Oloya et al 2006;OIE 2009). Indeed, Lesslie and Herbert (1975) and Lesslie et al (1975a, b), recorded hypersensitivity responses to AT that were equal to or higher than responses to BT in cattle naturally infected with M. bovis and presenting visible lesions at slaughter. The nature of the tuberculin is vital in non-specific skin test responses for the diagnosis of bovine TB, and the use of M. bovis-specific purified antigen as the test reagent is imperative for improved test performances (Pollock et al 2003;de la Rua-Domenech et al 2006a).…”

“…For bTB diagnosis, the single comparative intradermal tuberculin skin test (SCITT) using purified protein derivative tuberculin (PPD, Bovituber, Synbiotics Corporation, France) was performed on cattle as described by Lesslie & Herbert [32]. Three days post-injection, skin fold thicknesses were re-measured using callipers and positivity assessed according to Shirima et al [33].…”
