Comparison of the Digene HPV Genotyping LQ Test and the PANArray HPV Genotyping Chip for Detection of High-Risk or Probable High-Risk Human Papillomavirus Genotypes

Abstract: BackgroundWe evaluated the performance of two different array-based techniques, a bead-based multiplex genotyping method (LQ; digene HPV Genotyping LQ Test, QIAGEN, Germany) and a DNA chip-based method using peptide nucleic acid probes (PANArray; PANArray HPV Genotyping Chip, Panagene, Korea), for detection of human papillomavirus (HPV) and genotyping of high-risk (HR) or probable high-risk (PHR) HPVs in healthy patients who visited a health-promotion center.MethodsWe obtained 508 unselected, consecutive cervi… Show more

“…There have been conflicting results about the PCR-based DNA chip method in previous studies in Korea. The majority of these studies reported that the HPV DNA chip method had higher or similar sensitivity or detection rates, compared to HC2 or other DNA tests [ 21 - 24 ], whereas some reports showed that commercial DNA chips had lower sensitivity or detection rates for HR-HPV [ 25 , 26 ]. Specifically, there were contradictory reports about comparative results of the PANArray method in the respective studies [ 24 , 26 ].…”

“…In one report regarding human papillomavirus (HPV) detection, a microspherebased multiplex genotyping method (digene HPV Genotyping LQ Test, QIAGEN, Germany) showed higher concordance than the results of using PNA-based methods (PANArray HPV Genotyping Chip, Panagene, Korea) with a pool of 508 clinical specimens (Park et al 2014). This report also verified that the microsphere-based genotyping method is superior to sequencing when determining multiple HPV genotypes existing in one subject, since low concentrations of DNA may lead to less sequencing accuracy (Park et al 2014).…”

In another study, Luminex Mebgen HPV assay was considered superior compared with other commercially available HPV tests such as Hybrid Capture ™ 2 (HC2) test from Qiagen, the Linear Array ™ HPV genotyping test/line-blot assay and the Amplicor ™ HPV test from Roche (Ozaki et al 2014).…”
