Comparison of Stratum Corneum and Hair Fibrous Proteins

Abstract: The Tris-urea-mercaptoethanol (pH 9) soluble fibrous proteins of hair and stratum corneum have been compared in a number of animals. The 2 tissues show different urea (pH 8.3) and SDS polyacrylamide gel electrophoretic patterns and vary in amino acid composition. Cyanogen bromide cleavage was done on human stratum corneum and hair fibrous proteins and found to give different fragments by electrophoresis. Antibodies to stratum corneum protein did not react with hair protein and vice versa. Although the 2 types … Show more

“…but also to that reported for the nor mal human hair [5,19], This suggests that in the present study the hair SCM fibrous protein preparations analyzed were properly prepared. The amino acid composition of the HN-4' component from the patient was very similar to that of the HN-4 component from the normal individual (table 1).…”

“…Proteins were extracted from a hair specimen with 0.2 M Tris-HCl (pH 9.5) containing 8 M urea and 0.2 M 2-mercaptoethanol (5,15]. After centrifugation, the supernatant obtained was treated with iodoacetic acid to yield the S-carboxymcthylated (SCM) derivatives.…”

“…To define the optimal amino acid combination to use, we analyzed human skin scales by HPLC. The amino acid composition of human scales was found to compare with data available from the literature, as it relates to the human stratum corneum[35, 36] (data not shown). On the basis of this analysis, for the subsequent cultures we selected a mix of amino acid referred to as ‘synthetic aa mix’ (see ‘Materials and Methods’), commonly used in humans for parenteral nutrition.…”

“…Exactly 100 mg of the washed specimen as weighed with an electric balance (model PC2000, Mettler Instrumente AG, Zurich) was suspended in 100 ml of a freshly prepared solution of 0.2M tris-HCI (pH 9.5), 8M urea, and 0.2M 2-mercaptoethanol (4,5). It was then stirred at 100rpm under nitrogen at 50°Cfor 1 hr in a water incubator (model M-I00 N , Taiyo Science Industrial Co., Tokyo).…”
