Comparison of Seven Cell Lines Derived From Human Gastric Carcinomas

Abstract: In an attempt to elucidate various histological features of gastric cancers, seven human gastric adenocarcinomas were studied in vitro and in nude mice. Growth pattern of each cultured cell line in vitro corresponded well to the histological type of parent tumor. The cell lines, MKN7, MKN74, and MKN28 derived from differentiated carcinomas showed morphological characteristics of intestinal differentiation in cell polarity and microvilli with core-filaments in vitro as well as in nude mice. However, they gradua… Show more

“…33 OCUM and fibroblast cell lines were routinely kept in high glucose DMEM (4.5 g/l) with 10% FBS, 100 U/ml penicillin, 100 mg/ml streptomycin, 0.5 mM sodium pyruvate and 2 mM L-glutamine at 37 1C in 5% CO 2 (everything PAA Laboratories, Pasching, Austria). MKN-45 cells 34 were cultured according to the protocol outlined by the provider (German Collection of Microorganisms and Cell Cultures).…”

“…31,32,34 In vitro, OCUM cells exclusively grow in suspension (single or in cell clusters), whereas MKN-45 cells predominantly grow adherent as mono or multilayers with only few cells being suspended. This behavior is not characteristic for diffuse-type gastric tumors or their cell lines.…”

“…Ten gastric cancer cell lines with di ering histologies were cultured under appropriate conditions in our laboratory; MKN1, an adenosquamous cell carcinoma; MKN7, a well di erentiated adenocarcinoma; MKN28 and MKN74, moderately di erentiated adenocarcinomas, MKN45 and KWS-I; poorly di erentiated adenocarcinomas; KATO-III, a signet ring cell carcinoma; ECC10 and ECC12, endocrine cell carcinomas; and TSG11, a hepatoid carcinoma (Motoyama et al, 1986). Forty gastric cancers and their matching normal gastric mucosae were obtained surgically from 40 Japanese patients.…”
