Comparison of RNA In Situ Hybridization and Immunohistochemistry Techniques for the Detection and Localization of SARS-CoV-2 in Human Tissues

Abstract: Coronavirus disease-19 (COVID-19) is caused by a newly discovered coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Although SARS-CoV-2 is visualized on electron microscopy, there is an increasing demand for widely applicable techniques to visualize viral components within tissue specimens. Viral protein and RNA can be detected on formalin-fixed paraffin-embedded (FFPE) tissue using immunohistochemistry (IHC) and in situ hybridization (ISH), respectively. Herein, we evaluate the staini… Show more

“…Discrepant viral evidence results between qRT-PCR, ISH and IF/IHC technologies have been the subject of several recent studies 60 , 61 , 62 , 63 . The presence of current viral load, the phase of disease course as well as the severity of COVID-19 all have impact on the detection rate of viral RNA or viral protein in a given sample 60 , 61 , 62 , 63 .…”

“…Discrepancies between qRT-PCR and ISH RNA results (ISH negative and qRT-PCR positive pulmonary samples) were demonstrated on FFPE tissue in a large autopsy study by Massoth et al. [64] . Another recent study by Ko et al.…”

“…Another important concept to keep in mind is that viral components (RNA and proteins) are not produced in balanced amounts (as suggested in Figure 1 and Massoth et al 113 ). Therefore, surplus viral RNA and proteins may be encountered at the site of infection, in the circulation and at distant sites.…”

“…[4][5][6][7][8][9] Although evidence suggests that COVID-19 also impacts other organs such as heart 9 , liver 9 , and kidneys 9,10 , the virus has so far only been identified with certainty in the lungs and only rarely the brain, heart, liver, kidney, placenta, and blood by electron microscopy, immunohistochemistry (IHC), in situ hybridization (ISH), and reverse transcriptionpolymerase chain reaction (RT-PCR). 8,9,[11][12][13][14][15][16][17][18] SARS-CoV-2 infection is usually identified by the detection of viral RNA using RT-PCR on nasopharyngeal or oropharyngeal swabs. Current or prior infection might also be recognized by a serum test for SARS-CoV-2 IgG antibodies.…”
