Comparison of radiolabeled choline and ethanolamine as probe for cancer detection

Abstract: Methods: We carried out comparison of radiolabeled choline and ethanolamine by cell uptake study using 8 different cancer cell lines. To evaluate the response of radiotracers towards proliferation we also carried out their cell uptake study of androgen dependent and androgen independent cells in androgen stipulated and deprived media. Conclusions: Our data demonstrate that ethanolamine and N, N'-dimethyl ethanolamine are taken up by a wide variety of tumor cells significantly better (2-7 fold) than the clinical… Show more

“…As reported for other matrices, metabolomics studies in PCa also revealed that PCa cells experience alterations in membrane metabolism with changes, for example, in choline metabolite levels (Table 5) [98], [101]. Androgen signaling has an important role in the development and progression of PCa.…”

“…Androgen-nonresponsive cell lines and androgen-responsive cell lines also have differences in their glycolytic metabolism profiles. Androgen-dependent PCa cells and androgen-independent PCa cells also show differences in membrane lipid synthesis (Table 5) [97], [100], [101].…”

“…ethanolamine labelled with 11 C or 18 F) is a potential new probe in oncology PET-CT imaging for assessment of tumour proliferation [93]. 14 C-ethanolamine has been used in a variety of tumour cell types (i.e.…”

“…21 However, given the higher abundance of PE in plasma membranes, it was only a matter of time before the uptake of ethanolamine was determined in tumor cells. In the current issue of Cancer Biology and Therapy, Mintz et al 22 evaluate the uptake and retention of ethanolamine, N-N dimethylethanolamine and choline in a variety of tumor cell lines, including prostate and cerebral tumors. The authors show that across the spectrum of cell lines tested, tumor cells concentrated ethanolamine 2 to 7 fold higher than choline when incubated with these tracers.…”

“…Stimulation of prostate cancer cell proliferation with androgens enhanced uptake of the labeled substrates even further. Given the recent success with 11 C-choline imaging of tumors 20,21 and the results reported by Mintz et al 22 , it is clear that the next step will be in vivo studies with tumor xenografts to directly compare the two radiolabels in a more clinically relevant setting. The half-life of 11 C (20.38 minutes) may limit the availability of radiotracers based on this isotope and the longer path length of the emitted positron compared to that from 18 F can have a negative impact on image resolution.…”
