Comparison of Potentials of Stem Cells Isolated from Tendon and Bone Marrow for Musculoskeletal Tissue Engineering

Abstract: The use of tendon-derived stem cells (TDSCs) as a cell source for musculoskeletal tissue engineering has not been compared with that of bone marrow stromal cells (BMSC). This study compared the mesenchymal stem cell (MSC) and embryonic stem cells (ESC) markers, clonogenicity, proliferative capacity, and multilineage differentiation potential of rat TDSC and BMSC in vitro. The MSC and ESC marker profiles of paired TDSC and BMSC were compared using flow cytometry and quantitative real-time polymerase chain react… Show more

“…As in equine MSCs, CD 29 and CD 44 expressions are found consistently in canine (34,35,51,52), ovine (53), and porcine (54) MSCs. Corresponding to the equine antigen expression profile as well, CD 73 could not be detected in canine, rabbit, or sheep MSCs (35,55,56), while rat tendon derived, feline and bovine neonatal cell sources and caprine bone marrow-derived MSCs expressed this antigen (57)(58)(59)(60). CD 73 protein expression could also be detected in bone marrow derived MSCs from mice but only in moderate amounts (61).…”

“…5,33 Along this line, a very recent study reported an in-depth comparison of the phenotype and the regenerative potential of rat TDSCs and BMSCs. 31 Interestingly, the authors found that TDSCs possess a higher proliferative rate and multilineage differentiation potential than BMSCs, therefore concluding that TDSCs are a very promising stem cell source for musculoskeletal tissue engineering.…”

“…Spontaneous osteogenesis was also reported in other mesenchymal stem cells (MSCs), such as BMSCs [26,27], which might be due to endogenous BMP expression in MSCs [28]. Our previous study showed that TDSCs also exhibited higher osteogenic differentiation potential in osteogenic medium supplemented with dexamethosone, β-glycerolphosphate and ascorbic acid compared with BMSCs isolated from the same animal [29]. Our results thus suggest that TDSCs might be an attractive source for TBJ repair upon endogenous and/or exogenous BMP-2 stimulation, as bone formation is one of the key determinants of successful TBJ regeneration.…”
