Comparison of lansoprazole and omeprazole in the short‐term management of duodenal ulcers in Taiwan

Abstract: Lansoprazole is a novel H+/K+-ATPase inhibitor which exhibits a very potent and long-acting suppression of acid secretion. The object of this randomized, controlled trial was to compare duodenal ulcer healing rates after 4 weeks of treatment with a once-daily regimen of either 30 mg lansoprazole or 20 mg omeprazole. Of a total of 111 patients 57 received lansoprazole, whereas 54 received omeprazole. In intention-to-treat analysis at 4 weeks, 89.5% patients showed ulcer healing following treatment with lansopra… Show more

“…Diarrhoea, and not constipation, caused by diminishing chloride output from the stomach and a reduction of faecal water (as in congenital chloride diarrhoea; Aichbichler 1997) is also usually observed as side‐effect. However, other studies have suggested that constipation is increased or worsened after starting PPIs (Chang et al 1995; Garrigues et al 1996). A systemic testing of bowel function in the population with colonic manometry and the influence of drugs has not yet been performed.…”

“…Short-term daily omeprazole consumption delays solid GE in normal subjects [24][25][26][27][28]. On the other hand, lansoprazole, another proton pump inhibitor showing similar ulcer healing ability, has no effect on solid GE [29,30]. Perhaps variations in GE measurement, test meal, period of omeprazole premedication and studied subjects may account for these contradictory results.…”

“…Several studies have compared lansoprazole with omeprazole in the treatment of acute duodenal ulcer 82 –86 . None of these studies have shown a statistically significant difference in healing rates at 4 weeks, although in two studies lansoprazole 30 mg was associated with an increased healing rate at 2 weeks compared with omeprazole 20 mg 85 –86 .…”
